Summary: EXECUTIVE SUMMARY: DNA Extractionis the isolation of Deoxyribonucleic Acid (DNA) from plant, animal, bacteria and viruses which it commonly resides and it is basically used in medical examinations, clinical diagnostics, and forensic investigation through DNA finger printing. In this paper, DNA extraction was limited to classroom-based, junior level science activity leading to the development of the procedure fitted to one-hour class period. The study entitled "Development of Classroom-Based DNA Extraction Activity for Junior High School", aimed to lessen the time of extraction by identifying the materials needed to extract genomic DNA from plants using the conventional method or protocol applied in the classroom but utilized powdered unripe papaya fruit and powdered pineapple peel instead of meat tenderizer. The extraction was compared with commercial standard bromelain and papain in terms of extraction. Extraction time recorded exempted the preparation of materials and preparing the mixture to be used in extraction. The developed procedure for classroom-based DNA Extraction applied several modifications in the conventional standard procedure used in extracting genomic DNA. These were (1) skipping the hot bath for the mixture; (2) using the cold water bath for alcohol; (3) using 70% Isopropyl alcohol instead of 90-99%; (4) using banana and tomato instead of strawberry; (5) using powdered pineapple peel and powdered unripe papaya fruit with bromelain and papain. Moreover, the developed DNA Extraction Kit was composed of the DNA extraction activity sheet, the materials needed for extraction to include measuring cups, test tubes, stirrers, micro spatula, zip lock and mixing container, 6.25 grams and 25 grams powdered pineapple peel and powdered unripe papaya fruit. Findings described a comparable extraction time using powdered pineapple peel and powdered unripe papaya fruit with reference to commercial standard bromelain and papain enzyme. The enzymes function to speed up DNA extraction time. Further, powdered pineapple peel yielded comparable genomic DNA in a shorter time as compared with meat tenderizer using all raw materials; powdered unripe papaya fruit also yielded comparable DNA in lesser time in two raw materials and more time with a difference of 0.02 minutes in favor of meat tenderizer. Further, green luquid detergent was recommended over blue and yellow liquid detergent for faster rate of extraction; 6.25 grams of powdered unripe papaya fruit in extracting genomic DNA from tomato and banana; 25 grams in extracting DNA from pea; 25 grams of powdered pineapple peel in extracting genomic DNA from tomato and banana: and 0.5 grams in extracting DNA from pea. However, parallel study was recommended on using raw materials other than plants in extracting genomic DNA and on testing the validity and acceptability of the developed procedure to testing the effectiveness of the DNA extraction activity in the junior high school biology class.
