EXECUTIVE SUMMARY: The Study aimed to find the antimutagenic activity of Lipote fruit extract and to explore the biological activity and utilization of this fruit that may encourage propagation of this endangered Philippine endemic plant to avoid the possible extinction. The study was conducted from November 2014 to October 2015 in University of Rizal System Morong, Rizal. Experimental research design was employed in the study. The extraction of Lipote fruit was done at Room 311, Isidro S. Rodriguez building of University of Rizal System Morong Campus. The antimutagenecity test was conducted at Natural Product Chemistry Department-Institute of Chemistry research building National Science Complex, University of the Philippines, Diliman Quezon City, Metro Manila. To determine the antimutagenic activity of Lipote fruit extract, Ames test was used. It is a test that is widely used to identify the mutagenic potential of drugs, chemicals as well as plant materials and their direct and indirect mutagenic potential. The test organism used is Salmonella typhimurium TA100, since it carries mutation in genes involved in histidine synthesis and is used to detect frameshift mutation. The cells of test strain in the absence of an external histidine source cannot grow to form colonies thus, a colony growth resumed if a reversion of the mutation occurs allowing the production of histidine to resume. In determining the rate of the number of revertant colony forming units (CFU) on positive control plate, spontaneous control plate and the experiment plate, mean and standard deviation was used. The antimutagenic activity of Lipote fruit extract against S. typhimurium Ta100, determined by calculating the percent inhibitory activity. The result shows that, the lipote fruit extract has strong inhibitory activity against Salmonella typhimurium (TA100). Considering the low number of mutants in the experimental plate. it shows that, Lipote fruit extract has antimutagenic activity. Based from the findings of the study, it is recommended to utilize Lipote fruit as food supplement and a source of antioxidant.
